Intended Use

This kit is to detect Mycobacterium tuberculosis specific T cell immune response in lymphocytes isolated from human fresh peripheral venous anticoagulant whole blood.



Principle of the Procedure

Tubercul-specific T cells are present  in the peripheral blood  of mycobacterium  tuberculosis infected persons. After being stimulated by tubercul-specific proteins, these cells are activated and proliferated rapidly to secrete tubercul-related cytokines.

This kit selected ESAT-6, CFP-10, Rvl985c proteins encoded by RD1 and RD2 regions, which are present in  Mycobacterium tuberculosis  but are generally absent in BCG and most  non- tuberculosis mycobacteria.  It was expressed as fusion protein (ESAT-6-CFP-10-Rvl985c)  by genetic engineering technology. During the detection, the human peripheral blood mononuclear cells were isolated from the whole blood sample to eliminate blood background interference factors, and the influence of individual differences in the number of immune cells in the population was excluded by counting the number of mononuclear cells. Quantitative mononuclear cells were co-cultured with fusion protein ESAT-6-CFP-10-Rv1985c on a cell culture plate. Tubercul-specific T cells secreted Interferon-γ and interleukin-2 factors due to memory response. The concentrations of Interferon-γ and interleukin-2 in the culture supernatant were detected by double-antibody sandwich ELISA. To determine whether there is a mycobacterium tuberculosis specific cellular immune response. This method is not affected by the BCG vaccine and specifically detects tuberculosis caused by Mycobacterium tuberculosis.
